چکیده

Introduction Platelet-rich-plasma (PRP) is autologous plasma that has a platelet concentration above baseline. Upon activation, PRP releases a high concentration of various growth factors. Platelet -released growth factors (PRGF) have been used to support the repair of bone defects, especially in maxillofacial and dental surgery, yet their exact effect on bone healing is not clear. We previously showed a significant osteoinductive effect of PRGF on Mesenchymal Stem Cells (MSC), as shown by an increased expression of typical osteoblastic marker genes and matrix mineralization. In particular, we showed that PRGF up-regulates BMP2 at both gene and protein level. Furthermore, our observations suggested the possible involvement of the Wnt-signalling cascade. In the present work, we further investigate the intracellular pathways leading to MSCs osteogenic differentiation under PRGF stimulation, compared to the classical osteogenic media (Dexamethasone and BMP2 containing media).

بررسی الگوی متیلاسیون و بیان ژن APC به دنبال تمایز سلول‌های بنیادی مزانشیمی مشتق از مغز استخوان به رده استئوبلاستی

Background and Aim: Different regulation processes have an effect on osteoblastic differentiation of mesenchymal stem cells (MSCs), and among them Wnt signaling pathway is particularly desirable. In Wnt signaling pathway, Adenomatous Polyposis Coli (APC) bind to β-catenin and induce its degradation, thereby acting as a negative regulator of canonical Wnt pathway. In this study, gene expression ...

WNT3A modulates chondrogenesis via canonical and non-canonical Wnt pathways in MSCs.

The multilineage commitment of mesenchymal stem cells (MSCs) is controlled via unknown mechanisms. In this study, we investigated the regulation of the differentiation of MSCs into chondrocytes via the Wnt signaling pathway. Overexpression of WNT3A in MSCs activated both the canonical and non-canonical Wnt pathways, which were responsible for different WNT3A-induced outcomes. WNT3A promoted MSC...

Correction: TGF-β Prevents Phosphate-Induced Osteogenesis through Inhibition of BMP and Wnt/β-Catenin Pathways

BACKGROUND Transforming growth factor-β (TGF-β) is a key cytokine during differentiation of mesenchymal stem cells (MSC) into vascular smooth muscle cells (VSMC). High phosphate induces a phenotypic transformation of vascular smooth muscle cells (VSMC) into osteogenic-like cells. This study was aimed to evaluate signaling pathways involved during VSMC differentiation of MSC in presence or not o...

Prostate cancer induces bone metastasis through Wnt-induced bone morphogenetic protein-dependent and independent mechanisms.

Prostate cancer (PCa) is frequently accompanied by osteosclerotic (i.e., excessive bone production) bone metastases. Although bone morphogenetic proteins (BMP) and Wnts are mediators of PCa-induced osteoblastic activity, the relation between them in PCa bone metastases is unknown. The goal of this study was to define this relationship. Wnt3a and Wnt5a administration or knockdown of DKK-1, a Wnt...

Platelet-released supernatant induces osteoblastic differentiation of human mesenchymal stem cells: potential role of BMP-2.

Platelet-rich preparations have recently gained popularity in maxillofacial and dental surgery, but their beneficial effect is still under debate. Furthermore, very little is known about the effect of platelet preparations at the cellular level, and the underlying mechanisms. In this study, we tested the effect of platelet-released supernatant (PRS) on human mesenchymal stem cell (MSC) differen...
